摘 要:
在以‘阿蒂擎天’凤梨乙烯处理和不处理的植株为材料建立的抑制性差减杂交文库中, 筛选到的一个被乙烯诱导并与 已知植物钾转运体(potassium transporter)同源的cDNA片段, 通过RACE技术得到该基因的全长cDNA序列。生物信息学分 析表明, 该基因可能属于K+转运体超家族中的成员。利用半定量RT-PCR检测该基因的表达量在乙烯处理6 h会显著升高, 然后下降, 推测其受乙烯信号诱导, 并在后续一系列生理生化变化中发挥作用。关键词:钾转运体; ‘阿蒂擎天’; 乙烯; 克隆; 表达分析

Abstract:
A cDNA fragment homologue of the known plant potassium transporter was screened in the suppression subtractive hybridization (SSH) library with ethylene treated and untreated Guzmania wittmackii ‘Attila’ plants as the material. Rapid Amplification of cDNA Ends (RACE) was used to obtain the full length cDNA. Bioinformatics analysis results suggested that the gene belongs to K+ trans superfamily. Semi-quantitative RTPCR results indicated that the expression of this gene increased significantly in 6 h and then decreased after ethylene treatment, which suggested it could be induced by ethylene signal, and may play an important role in the subsequent physiological and biochemical processes.Key words: potassium transporter; Guzmania wittmackii ‘Attila’; ethylene; cloning; expression analysis
